What It Means

UHPLC-MS stands for Ultra-High-Performance Liquid Chromatography–Mass Spectrometry.

It combines two analytical techniques:

  • UHPLC separates the different chemicals present in a sample.
  • Mass Spectrometry (MS) identifies those chemicals and measures their amount.

So, in simple terms, UHPLC separates the mixture, while MS tells us what each separated molecule is.

How It Works

The sample, usually in liquid form, is passed through a special UHPLC column under very high pressure.

Different molecules move through the column at different speeds because they interact differently with the material inside it. This allows them to separate from one another.

After separation, the molecules enter the mass spectrometer, where they are:

  • converted into charged particles called ions;
  • separated according to their mass-to-charge ratio;
  • detected and identified.

Compared with conventional HPLC, UHPLC uses smaller particles and higher pressure, giving:

  • faster analysis;
  • sharper separation;
  • better sensitivity;
  • improved detection of substances present in very small amounts.

Applications

UHPLC-MS is widely used in:

  • metabolomics;
  • pharmaceutical research;
  • drug and toxin detection;
  • food-safety testing;
  • pesticide analysis;
  • environmental monitoring;
  • clinical research;
  • forensic science.

In metabolomics, for example, UHPLC-MS can separate and measure hundreds or even thousands of metabolites present in blood, urine or tissue.

A more advanced form, UHPLC-MS/MS, analyses a molecule and then breaks it into smaller fragments. These fragments help confirm the molecule’s identity more accurately.

Its major strength is the ability to analyse complex mixtures quickly and with very high sensitivity. The main limitations are expensive equipment, specialised expertise and the large amount of data produced.

UHPLC-MS is therefore a powerful technique for separating, identifying and measuring many chemicals in a single complex sample.
